Neurotransmission of cognition, part 1, Dopamine is a hitchhiker in frontal cortex: norepinephrine transporters regulate dopamine.

چکیده

ISSUE Since the frontal cortex has a low density of dopamine transporters, dopamine has to be inactivated there by hitching a ride on the norepinephrine transporter of neighboring norepinephrine neurons.

Neurotransmission of cognition, part 3. Mechanism of action of selective NRIs: both dopamine and norepinephrine increase in prefrontal cortex.

ISSUE Selective norepinephrine reuptake inhibitors exploit the fact that dopamine transporters are absent in prefrontal cortex, so dopamine has to hitchhike a ride on the norepinephrine transporter in order to be inactivated. Thus, blocking norepinephrine transporters leads to an increase in both dopamine and norepinephrine levels in prefrontal cortex as well as improvement in cognition in atte...

Neurotransmission of cognition, part 2. Selective NRIs are smart drugs: exploiting regionally selective actions on both dopamine and norepinephrine to enhance cognition.

ISSUE Selective norepinephrine reuptake inhibitors such as atomoxetine increase both dopamine and norepinephrine in frontal cortex and may thereby enhance cognitive functioning in attention-deficit/hyperactivity disorder.

Dopamine uptake through the norepinephrine transporter in brain regions with low levels of the dopamine transporter: evidence from knock-out mouse lines.

Selective blockers of the norepinephrine transporter (NET) inhibit dopamine uptake in the prefrontal cortex. This suggests that dopamine in this region is normally cleared by the somewhat promiscuous NET. We have tested this hypothesis by comparing the effects of inhibitors selective for the three monoamine transporters with those of a nonspecific inhibitor, cocaine, on uptake of 3H-dopamine in...

Handbook of Neurochemistry and Molecular Neurobiology: Schizophrenia

This chapter is a review of the evidence implicating a dysfunction of dopamine transmission inthe prefrontal cortex in schizophrenia, underlying negative symptoms and cognitive deficits. The evidencederives essentially from brain imaging studies, post-mortem studies and clinical pharmacological studies. The imaging studies have suggested alterations in the main mediator of dopamine t...

Novel mechanism of antidepressant action: norepinephrine and dopamine disinhibition (NDDI) plus melatonergic agonism.

All known antidepressants enhance monoamine neurotransmission, and most do so by blocking one or more of the reuptake transporters for serotonin, norepinephrine and/or dopamine (Stahl, 2000). Now comes agomelatine, a new agent that enhances norepinephrine and dopamine neurotransmission by a unique mechanism, namely 5-HT2C antagonism (Millan et al., 2003; Millan, 2005). Agomelatine is also a dir...
